    There must be reasons why your wife is concerned. Though you have placed yourself on a defemse mode, implies you too must inward aware you are wee... bit too too close.
    Identify those reasons, eg, being too cosy, jokes, suddenly being animated in the poresence of your lady colleague, or sharing snacks, lunch moments, and spending too much together during breaks, walks, or exchanging snacks, phionefall at family time, after hours, gettinh involved in your collegaues personal issues, including lifts and picking groceries etc.
    Iy would be unwise to loose your wife for a job, nor would leaving your job - solve problem.
    Perhaps you are a nice guy hence will carry same behaviour.
    Recommendation: If you love, care about your wife, you have to re-assure her by keeping healthy boundaries with your colleague. That require you have a candid and honest converstion with your colleague, share your concerns and cut off some tell tale signs that has created that impression that you are disturbingly too close.
    Your family well being matters more than anything.
