1) Project: Development of a rapid sequencing method for adventitious virus detection to monitor CAR-T cells' quality and safety attributes.
Responsibilities:
Reviewed literature on next-generation sequencing to identify methods relevant to the project.
Designed, executed, and troubleshot virus spiking experiments to develop a rapid method for contaminant virus detection using long read sequencers. We have submitted two invention disclosures in relation to sample pre-treatment and pre-processing.
Prepared sequencing libraries, operated the GriDION sequencing device, analyzed, and interpreted genomic data.
Presented results during internal group meetings and relevant conferences such as (a) the 2022 conference on Next Generation Sequencing organized by the International Alliance for Biological Standardization and (b) the 2022 MIT BioMANN summit.
Mentored undergraduate students on molecular biology research techniques and procedures.
Ordered and managed laboratory equipment and supplies.
Recorded laboratory data in electronic notebook (LabArchives) and authored Standard Operating Protocols to ensure reproducibility of the developed method.
2) Collaborative works:
Collaborated (a) with industry and academic scientists in efforts to develop a next-generation sequencing method as part of the ADVentitious agent Detection Technology Interest Group (ADVTIG) (b) with the lead scientists to write grants; (c) with electrical engineers to evaluate the performance of virus concentration devices for enhanced virus detection.
WHO International Virus Panel Contribution
Successfully sequenced and analyzed virus genomic data from a multiplex viral reference reagent,
Provided critical data from Oxford Nanopore Sequencing contributing to a comprehensive dataset,
The collective data generated was instrumental in establishing the 1st WHO International virus panel for adventitious virus detection in biological products.
